چکیده
Autophagy contributes to sebaceous gland function and the control of sebum composition. Based on literature data, autophagy markers were strongly expressed in maturing cells healthy skin, whereas downregulated acne-involved glands. Interestingly, tissue transglutaminase (TG2) which is known regulate mouse embryonic fibroblast has a potential role also integrity epidermis exerts an abundant expression In this work therefore, our aim was assess TG2 biology. Using knockout (KO) mice, water retention assay showed that repulsion higher KO mice compared wild-type animals. While Western blot high-content screening (HCS) results using permanent SZ95 sebocyte cell line generated with CRISPR/Cas9 method, absence sebocytes accumulated lipid droplets amounts, phenomenon associated lower levels such as ATG5, ATG7, LC3II/I. Our suggest may contribute production via regulation thus add TG2-related regulatory processes list possible targets for interventions dermatological disorders altered production.

Particulate matter 2.5 regulates lipid synthesis and inflammatory cytokine production in human SZ95 sebocytes
A large body of evidence indicates that particulate matter (PM)2.5 is associated with various negative effects on human health. However, the impact and molecular mechanism of PM2.5 on the skin have not been elucidated. Therefore, the present study aimed to investigate the effects of two types of PM2.5 [water-soluble extracts (W-PM2.5) and non-water-soluble extracts (NW-PM2.5)] on cell prolifera...
متن کاملAutophagy deficiency leads to accumulation of ubiquitinated proteins, ER stress, and cell death in Arabidopsis
Autophagy is a homeostatic degradation and recycling process that is also involved in defense against microbial pathogens and in certain forms of cellular suicide. Autophagy has been proposed to negatively regulate plant immunity-associated cell death related to the hypersensitive response (HR), as older autophagy-deficient mutants are unable to contain this type of cell death 5 to 10 d after i...
متن کاملArachidonate 15-Lipoxygenase Type B Knockdown Leads to Reduced Lipid Accumulation and Inflammation in Atherosclerosis
Inflammation in the vascular wall is important for development of atherosclerosis. We have shown previously that arachidonate 15-lipoxygenase type B (ALOX15B) is more highly expressed in human atherosclerotic lesions than in healthy arteries. This enzyme oxidizes fatty acids to substances that promote local inflammation and is expressed in lipid-loaded macrophages (foam cells) present in the at...
متن کاملLipid Peroxidation After Ionizing Irradiation Leads to Apoptosis and Autophagy
A living cell is a dynamic biological system composed primarily of nucleic acids, carbohydrates, lipids, and proteins that structurally and functionally interact with many other molecules--organic and inorganic--to carry out normal cell metabolism. Exposure of a cell to radiation can both directly and indirectly alter molecules within the cell to affect cell viability. Radiation energy absorbed...
متن کاملEther lipid biosynthesis: alkyl-dihydroxyacetonephosphate synthase protein deficiency leads to reduced dihydroxyacetonephosphate acyltransferase activities.
Recent studies have indicated that two peroxisomal enzymes involved in ether lipid synthesis, i.e., dihydroxyacetonephosphate acyltransferase and alkyl-dihydroxyacetonephosphate synthase, are directed to peroxisomes by different targeting signals, i.e., peroxisomal targeting signal type 1 and type 2, respectively. In this study, we describe a new human fibroblast cell line in which alkyl-dihydr...
